IGF1-LR3 — Long Arginine Insulin-Like Growth Factor
IGF1-LR3 (Long Arginine 3 IGF-1) is an 83-amino acid synthetic analog of insulin-like growth factor 1, featuring a 13-amino acid N-terminal extension and an arginine substitution at position 3. These modifications dramatically reduce binding to IGF binding proteins (IGFBPs), resulting in a compound with approximately 2-3x the potency and significantly longer functional half-life compared to native IGF-1.
Mechanism of Action
IGF1-LR3 binds to the IGF-1 receptor (IGF-1R), a transmembrane tyrosine kinase receptor that activates both the PI3K/Akt/mTOR pathway (promoting protein synthesis and cell survival) and the Ras/MAPK pathway (promoting cell proliferation and differentiation). Because LR3 has minimal affinity for IGFBPs, a greater proportion of the peptide remains bioactive in circulation, providing enhanced and prolonged receptor stimulation compared to recombinant human IGF-1.
